Abcam's DKK4 (Dickkopf-4) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human DKK4 in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for Human DKK4 coated on a 96-well plate. Standards and samples are pipetted into the wells and DKK4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human DKK4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DKK4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimisation may be required with urine samples.

Biological function summary
DKK4 participates in the regulation of embryonic development and tissue homeostasis. It functions as a part of the larger Dickkopf protein family which modulates Wnt signaling by binding to LRP5/6 co-receptors. Through this interaction DKK4 manages the balance between cell growth and apoptosis important in both early development and adult tissue maintenance. DKK4 presence is often necessary to modulate signals that govern differentiation and proliferation in complex tissue systems.
Pathways
DKK4 predominantly interacts within the Wnt signaling pathway. This pathway is critical for regulating gene expression involved in stem cell maintenance and differentiation. DKK4 acts by interacting with proteins such as LRP6 and Kremen thereby inhibiting Wnt-induced processes. Through modulation of these factors DKK4 ensures the fine-tuning of signaling cascades necessary for maintaining cellular equilibrium under physiological conditions.
