Interpolated concentrations of native DLK1 in human serum and plasma samples.
The concentrations of DLK1 were measured in duplicates, interpolated from the DLK1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5%, plasma (citrate) 25%, plasma (EDTA) 25%, and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DLK1 concentration was determined to be 10.36 ng/mL in serum, 8.41 ng/mL in plasma (citrate), 5.99 ng/mL in plasma (EDTA), and 10.26 ng/mL in plasma (heparin).

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DLK1 concentration was determined to be 7 ng/mL with a range of 0 – 13 ng/mL.

The concentrations of DLK1 were measured in duplicate and interpolated from the DLK1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DLK1 concentration was determined to be 1,087 pg/mL in placenta extract.

Human DLK1 ELISA Kit (ab270211) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of DLK1 protein in serum, cit plasma, hep plasma, tissue extracts, and edta plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human DLK1 with 35 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Protein delta homolog 1 (DLK1) is a protein that in humans, is encoded by the DLK1 gene. DLK1 is expressed as a transmembrane protein, but a soluble form cleaved by ADAM17 is active in inhibiting adipogenesis, the differentiation of pre-adipocytes into adipocytes. It is a member of the EGF-like family of homeotic proteins. DLK1 is found within the stromal cells in close contact to the vascular structure of placental villi, yolk sac, fetal liver, adrenal cortex and pancreas and in the beta cells of the islets of Langerhans in the adult pancreas. Found also in some forms of neuroendocrine lung tumor tissue. Human DLK1 shares 85% sequence homology with mouse and rat, 83% and 93% with bovine and monkey, respectively.

Biological function summary
DLK-1 serves a function in the inhibition of adipocyte differentiation and the maintenance of preadipocyte phenotype. It does not form part of a complex but interacts with membrane-bound receptors to mediate signaling pathways that impact tissue development. Researchers study its role in embryonic development and its downregulation is a significant factor during the differentiation of mesenchymal cells. Due to these interactions DLK-1 has a pivotal involvement in processes governing tissue regeneration and growth control.
Pathways
Better understanding of DLK-1 involves examining its interaction in the Notch signaling pathway. This important pathway related to cell fate decisions features pathways with other proteins like Notch-1 and Notch-3. The relationship helps modulate the fate and proliferation of cells. DLK-1 also has associations with pathways involving phosphoinositide 3-kinase (PI3K) which influences cell growth and survival through related proteins such as AKT.
