Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean DNase I concentration was determined to be 4675 pg/mL with a range of 1271 – 12993 pg/mL.

Interpolated concentration of native DNase I was measured in duplicate at different sample concentrations. Undiluted samples are as follows : serum 25% plasma (citrate) 25% plasma (EDTA) 25% plasma (heparin) 25% and urine 1%.  The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD n=2). Sample dilutions are made in Sample Diluent NS.

How the assay works

Human DNase I ELISA Kit, Chemiluminescent (ab327311) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Product protocols

Target data

Serum endocuclease secreted into body fluids by a wide variety of exocrine and endocrine organs (PubMed : 11241278, PubMed : 2251263, PubMed : 2277032). Expressed by non-hematopoietic tissues and preferentially cleaves protein-free DNA (By similarity). Among other functions, seems to be involved in cell death by apoptosis (PubMed : 11241278). Binds specifically to G-actin and blocks actin polymerization (By similarity). Together with DNASE1L3, plays a key role in degrading neutrophil extracellular traps (NETs) (By similarity). NETs are mainly composed of DNA fibers and are released by neutrophils to bind pathogens during inflammation (By similarity). Degradation of intravascular NETs by DNASE1 and DNASE1L3 is required to prevent formation of clots that obstruct blood vessels and cause organ damage following inflammation (By similarity).
