Interpolated concentrations of native DR4 in human HeLa, K562 and HCT116 extracts, based on 500, 800 and 250 μg/mL extract loads, respectively, The concentrations of DR4 were measured in duplicate and interpolated from the DR4 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DR4 concentration was determined to be 119.90 pg/mL in HeLa extract, 169.71 pg/mL in K562 extract, and 397.09 pg/mL in HCT116 extract.

Human DR4 ELISA kit (ab282881) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human DR4 protein in human cell extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human DR4 with 5 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies

Human DR4 is a type 1, TNF R family, membrane protein. Human DR4 is a receptor for the cytotoxic ligand TNFSF10/TRAIL. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Human DR4 also promotes the activation of NF-kappa-B. DR4 is most closely related to DR5 sharing a 55% amino acid sequence identity.

Biological function summary
The death receptor 4 plays an important role in mediating cell apoptosis. Upon ligand binding DR4 interacts with the adapter molecule FADD (FAS-associated protein with death domain) facilitating the formation of the death-inducing signaling complex (DISC). This complex recruits and activates caspase-8 which eventually leads to the activation of the downstream effector caspases driving the cell towards apoptosis. DR4 operates as a significant component of the extrinsic pathway of apoptosis a process important for maintaining cellular homeostasis and eliminating harmful cells.
Pathways
DR4 functions critically within the extrinsic apoptotic pathway. This pathway involves other death receptors such as DR5 which also binds TRAIL similarly leading to apoptosis through DISC formation and caspase activation. The extrinsic pathway interconnects with the intrinsic pathway via the mitochondria with proteins like Bid acting as a bridge between them. The engagement of these pathways highlights DR4’s importance in regulatory mechanisms of cell death tightly controlling cellular proliferation and survival.
