Abcam's Human Endostatin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Endostatin in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for Human Endostatin coated on a 96-well plate. Standards and samples are pipetted into the wells and Endostatin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Endostatin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Endostatin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
Endostatin functions as an important inhibitor of angiogenesis the process in which new blood vessels form from pre-existing ones. It acts independently and is not part of a larger protein complex. In the cellular microenvironment endostatin binds to various integrins inhibiting endothelial cell proliferation and inducing apoptosis. It also disrupts signal transduction pathways critical for endothelial cell survival and migration.
Pathways
Endostatin holds an important position in the angiogenesis and Wnt signaling pathways. It interacts closely with proteins like VEGF (Vascular Endothelial Growth Factor) and FGF2 (Fibroblast Growth Factor 2) where it counteracts their pro-angiogenic effects.
