Abcam's anti-Entamoeba histolytica IgG Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgG class antibodies against Entamoeba histolytica in Human serum and plasma.
A 96-well plate has been precoated with Entamoeba histolytica antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a Protein A peroxidase labelled anti-Human IgG conjugate is added to the wells, which binds to the immobilized Entamoeba histolytica-specific antibodies. TMB is then catalyzed by the Protein A to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Entamoeba histolytica IgG sample captured in plate.

Biological function summary
Entamoeba histolytica IgG contributes to immune defense by identifying and marking Entamoeba histolytica cells for destruction by other immune cells. It does not form part of a larger protein complex but instead operates through its specific binding capacity. Upon binding to antigens presented by Entamoeba histolytica this IgG type facilitates the process of phagocytosis by engaging phagocytic cells such as macrophages and neutrophils. This immune response serves as a primary line of defense to control the infection and limit the spread of the parasite within the host's body.
Pathways
Entamoeba histolytica IgG gets engaged in the humoral immune response which is integral to the adaptive immune system. This process involves pathways like the complement system pathway wherein the binding of IgG to the parasite antigens activates a cascade that leads to opsonization complement fixation and eventual lysis of the parasitic cells. Additionally the antibody engages in antibody-dependent cellular cytotoxicity (ADCC) pathways which involve recruiting natural killer cells to target the infected cells thereby reducing parasite survival and proliferation.
