Sandwich ELISA - Human ErbB3 ELISA Kit (HER3) (AB275101)
Interpolated concentrations of native ErbB 3 in human serum, plasma and cell culture supernatant samples.
The concentrations of ErbB 3 were measured in duplicates, interpolated from the ErbB 3 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 25% and MCF-7 supernatant neat. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ErbB 3 concentration was determined to be 4,877 pg/mL in serum, 3,874 pg/mL in plasma (citrate), 4,148 pg/mL in plasma (EDTA), 3,783 pg/mL in plasma (heparin), and 301.2 pg/mL in MCF-7 supernatant.

Interpolated concentrations of native ErbB 3 in human MCF-7 based on an 800 μg/mL extract load.
The concentrations of ErbB 3 were measured in duplicate and interpolated from the ErbB 3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ErbB 3 concentration was determined to be 3,799 pg/mL in MCF-7 cell extract.

Product details
Human ErbB 3 ELISA kit (ab275101) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ErbB 3 protein in serum, plasma, cell culture supernatant, urine, saliva and cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ErbB 3 with 27.6 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Receptor tyrosine-protein kinase ErbB 3, also known human epidermal growth factor receptor 3 (HER3), is encoded by the ERBB3 gene in which is a member of the EGFR family. ErbB 3 is a type I transmembrane glycoprotein that binds to neuregulins NRG1 and NRG2, but lacks a working kinase domain. ErbB 3 is capable of heterodimerization of its fellow ErbB family members, most notably with ErbB 2 for signaling. Moreover, ErbB 3 is vital in the development of Schwann cells, neural crest differentiation, sympathetic nervous system, and heart values. Multiple studies have shown ErbB 3 to play roles in cellular growth, proliferation, differentiation, promotion and metastasis of cancer, and chemotherapeutic resistance.

Biological function summary
HER3 is essential for activating important signaling pathways that regulate proliferation differentiation and survival. The ErbB3 protein forms heterodimers with other ErbB family members such as ErbB2 (HER2) which is necessary for its signaling capability. The HER3 protein is abundant in heart and nervous tissues indicating its role in cardiovascular and neurological functions. Its activation modulates gene expression through downstream effectors mediated by dimerization.
Pathways
HER3 plays a pivotal role in the PI3K/Akt and MAPK signaling pathways. These pathways are fundamental in cellular responses like growth and survival. HER3 serves as a major partner for HER2 in these signaling cascades enhancing downstream signaling effects. HER3's connection to these pathways makes it a critical node collaborating closely with proteins such as HER2 and EGFR to maintain efficient communication and function within these networks.
