Sandwich ELISA - Human Ezrin ELISA Kit (AB278116)
Interpolated concentrations of native Ezrin in human serum and plasma samples.
The concentrations of Ezrin were measured in duplicates, interpolated from the Ezrin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 50%, plasma (heparin) 100% and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ezrin concentration was determined to be 3409.74 pg/mL in serum, 1833.59 pg/mL in plasma (citrate), 3172.31 pg/mL in plasma (EDTA), and 3470.33 pg/mL in plasma (heparin).

Product details
Human Ezrin ELISA kit (ab278116) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Ezrin protein in human serum and plasma samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Ezrin with 19.37 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Ezrin is a cytoplasmic peripheral protein involved in adhesion, migration, and organization of cytoskeletal structures to the plasma membrane. In epithelial cells, Ezrin is required for the formation of microvilli and membrane ruffles on the apical pole. The N-terminal domain of Ezrin binds to NHERF protein involved in long-range allostery. Along with PLEKHG6, Ezrin is required for normal macropinocytosis. Human Ezrin holds a 97% and 96% sequence homology with mouse and rat Ezrin, respectively.

Biological function summary
Ezrin functions as a membrane-microfilament linker and is essential for maintaining cell shape adhesion and motility. It is an integral component of many cellular structures including microvilli and ruffles. Ezrin’s activity often involves forming complexes with other proteins such as EBP50 and F-actin which are fundamental for its role in cellular morphogenesis. These interactions allow ezrin to regulate membrane surface structure and cell surface organization.
Pathways
Ezrin participates in various signaling pathways that are essential for cell migration and invasion. One key pathway is the Rho family of GTPases which regulates actin cytoskeleton dynamics. Ezrin interacts with proteins like Rho-associated kinase (ROCK) that influence cell contraction and motility. Additionally ezrin is involved in the MAPK/ERK pathway which influences cell growth and survival by interacting with other signaling molecules.
