Sandwich ELISA - Human FABP4 ELISA Kit (AB234565)
Interpolated concentrations of native FABP4 in human THP-1 cell supernatant, THP-1 cell extract, adipocyte cell extract, and heart extract.
The concentrations of FABP4 were measured in duplicate and interpolated from the FABP4 standard curve and corrected for sample dilution. Undiluted samples are as follows : THP-1 cell supernatant 50%, THP-1 cell extract 50 μg/mL, adipocyte cell extract 10 μg/mL, and heart extract 1,000 ng/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Sandwich ELISA - Human FABP4 ELISA Kit (AB234565)
Serum from ten individual healthy human female and male donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean FABP4 concentration was determined to be 18 ng/mL with a range of 7 – 33 ng/mL in the female donors and 12 ng/mL with a range of 8 – 15 ng/mL in the male donors.

Sandwich ELISA - Human FABP4 ELISA Kit (AB234565)
Interpolated concentrations of native FABP4 in human serum and plasma samples.
The concentrations of FABP4 were measured in duplicates, interpolated from the FABP4 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 80, plasma (heparin) 1 : 67, plasma (EDTA) 1 : 67, and plasma (citrate) 1 : 67. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean FABP4 concentration in the neat samples was determined to be 13 ng/mL in serum, 12 ng/mL in plasma (heparin), 7 ng/mL in plasma (EDTA), and 11 ng/mL in plasma (citrate).

Product details
Human FABP4 ELISA Kit (ab234565) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of FABP4 protein in cell culture extracts, cell culture supernatant, serum, tissue extracts, cit plasma, edta plasma, and hep plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human FABP4 with 2.7 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Biological function summary
This protein participates in the regulation of lipid homeostasis and energy metabolism. FABP4 is part of the lipid-binding protein family that helps maintain fatty acid equilibrium within cells. By controlling the transport and signaling of lipids FABP4 impacts lipid storage insulin sensitivity and inflammation processes. Its activity is not confined to a specific multimeric complex yet it interacts with other proteins to exert its functions contributing to significant metabolic pathways in the body.
Pathways
FABP4 plays essential roles in the regulation of metabolic and inflammatory signaling pathways. It is intricately involved in the peroxisome proliferator-activated receptor gamma (PPARγ) pathway where it modulates adipose tissue development and glucose metabolism. In addition FABP4 has connections with the c-Jun N-terminal kinase (JNK) signaling pathway influencing stress and inflammatory responses. Interactions with these pathways place FABP4 at a link with proteins like PPARγ and JNK essential for cellular metabolic balance and responses to metabolic stress.
