Interpolated concentrations of native Fetuin-A in human serum, plasma and cell culture supernatant samples.

The concentrations of Fetuin-A were measured in duplicates, interpolated from the Fetuin-A standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 20,000, plasma (citrate) 1 : 20,000, plasma (EDTA) 1 : 20,000, and HepG2 supernatant 1 : 600. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Fetuin-A concentration was determined to be 194.15 μg/mL in serum, 164.51 μg/mL in plasma (citrate), 172.44 μg/ml in plasma (EDTA), and 3.85 μg/mL HepG2 supernatant.

Human Fetuin-A ELISA Kit (ab269372) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fetuin-A protein in cell culture supernatant, edta plasma, serum, and cit plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Fetuin-A with 116.24 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Fetuin A is a highly abundant protein secreted by the liver that is capable of binding a wide variety of ligands, similar to serum albumin. A member of the cystatin family, Fetuin A plays a vital role in preventing tissue calcification and is also involved in fatty acid transport, insulin regulation and anti-inflammatory responses. Fetuin A is encoded by a single gene on chromosome 3, contains two cystatin domains, and exists as a heterodimer in its posttranslational form. Fetuin A is more abundant than serum albumin in fetal serum.

The target AHSG also known as fetuin-A or alpha-2-HS-glycoprotein is a multifunctional glycoprotein with an approximate mass of 64-68 kDa. It is mainly expressed in the liver and secreted into the bloodstream. AHSG exhibits a wide range of interactions with different cell types and biological molecules playing key roles in mineralization inhibition of arterial calcification and modulation of immune responses.
Biological function summary

AHSG protein serves multiple functions including acting as an inhibitor of insulin receptor autophosphorylation and serine-threonine kinase activities. AHSG is part of a complex dynamic process involving calcium metabolism and bone mineralization. It binds to transforming growth factor-beta (TGF-β) and other proteins modulating cellular signals and maintaining homeostasis in tissues.
