Acts as a ligand for FGFR1, FGFR2, FGFR3 and FGFR4 (PubMed:8663044). Also acts as an integrin ligand which is required for FGF2 signaling (PubMed:28302677). Binds to integrin ITGAV:ITGB3 (PubMed:28302677). Plays an important role in the regulation of cell survival, cell division, cell differentiation and cell migration (PubMed:28302677, PubMed:8663044). Functions as a potent mitogen in vitro (PubMed:1721615, PubMed:3732516, PubMed:3964259). Can induce angiogenesis (PubMed:23469107, PubMed:28302677). Abcam's FGF basic (FGF2) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human FGF basic in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for Human FGF basic coated on a 96-well plate. Standards and samples are pipetted into the wells and FGF basic present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human FGF basic antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FGF basic bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimization may be required with urine samples.
FGF basic also known as FGF-2 or bFGF is a member of the fibroblast growth factor family. This protein is approximately 18 kDa in mass and is expressed in various tissues including the brain skin and vascular endothelium. Its alternate forms include FGF-2 and FGF-basic which serve similar functional roles. FGF basic acts mechanically by binding to fibroblast growth factor receptors (FGFRs) initiating a cascade of intracellular signaling that leads to cell growth and differentiation.
Fibroblast growth factor 2 is important in development and wound healing. It does not form part of a larger complex but interacts directly with receptors on the cell surface to activate downstream signaling pathways. FGF basic plays an important role in angiogenesis facilitating the growth of new blood vessels. Additionally this protein supports the proliferation of a wide range of cell types including fibroblasts and endothelial cells.
Fibroblast growth factor 2 participates mainly in the MAPK/ERK and PI3K/AKT signaling pathways. These pathways are essential for controlling cell division and survival. In the MAPK/ERK pathway FGF basic's interaction with FGFR involves proteins such as Ras and Raf. Similarly in the PI3K/AKT pathway it engages with proteins including PI3K and Akt coordinating various cellular responses to environmental stimuli.
FGF basic links to various cancers and cardiovascular diseases. In cancer abnormal FGF basic signaling can lead to uncontrolled cell growth where it interacts with oncogenic proteins like Ras. In cardiovascular disorders altered FGF basic expression affects vascular integrity often in conjunction with other proteins like VEGF contributing to pathological conditions such as atherosclerosis.
