Together with fibrinogen alpha (FGA) and fibrinogen beta (FGB), polymerizes to form an insoluble fibrin matrix. Has a major function in hemostasis as one of the primary components of blood clots. In addition, functions during the early stages of wound repair to stabilize the lesion and guide cell migration during re-epithelialization. Was originally thought to be essential for platelet aggregation, based on in vitro studies using anticoagulated blood. However, subsequent studies have shown that it is not absolutely required for thrombus formation in vivo. Enhances expression of SELP in activated platelets via an ITGB3-dependent pathway. Maternal fibrinogen is essential for successful pregnancy. Fibrin deposition is also associated with infection, where it protects against IFNG-mediated hemorrhage. May also facilitate the antibacterial immune response via both innate and T-cell mediated pathways.

Human Fibrinogen gamma chain - Intracellular SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fibrinogen gamma chain - Intracellular protein in human serum, plasma, saliva and cell culture supernatant. Quantitate Human Fibrinogen gamma chain - Intracellular with 8.8 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (Pre-coated 384 well Microplate SimpleStep ELISA® ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
CROSSREACTIVITY
2,300 pg/mL of recombinant Human Fibrinogen alpha was tested for cross reactivity. No cross reactivity was observed.
2,300 pg/mL of recombinant Human Fibrinogen beta was tested for cross reactivity. <3% cross reactivity was observed.
INTERFERENCE
575 pg/mL of recombinant Human Fibrinogen alpha and 575 pg/mL of recombinant Human Fibrinogen beta was tested for interference with 575 pg/mL of recombinant Human Fibrinogen gamma. No interference was observed. 575 pg/mL of recombinant Human Fibrinogen beta increased interpolated value of 575 pg/mL of recombinant Human Fibrinogen gamma 5.7%.
SPECIES REACTIVITY
Other species reactivity was determined by measuring 1:1000 serum samples of various species, interpolating the protein concentrations from the species standard curve, and expressing the interpolated concentrations as a percentage of the protein concentration in species serum assayed at the same dilution.
Reactivity of 77% was determined for primate serum.
No signal was observed in 1:1000 serum samples from the following species: Mouse, Rat, Bovine
Other species reactivity not determined.

Example of Human Fibrinogen gamma chain standard curve. Background-subtracted data values (mean +/- SD) are graphed
Interpolated concentration of native Fibrinogen gamma chain was measured in duplicate at different sample concentrations. Undiluted samples are as follows: HepG2 Cell Extract 15 µg/mL, and Human Liver Tissue Extract 500 ng/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1x Cell Extraction Buffer PTR
