Interpolated concentrations of native Ficolin-2 in human serum and plasma samples.

The concentrations of Ficolin-2 were measured in duplicates, interpolated from the Ficolin-2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 200, plasma (citrate) 1 : 200, plasma (EDTA) 1 : 400, and plasma (heparin) 1 : 200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ficolin-2 concentration was determined to be 481.67 ng/mL in serum, 508.04 ng/mL in plasma (citrate), 443.89 ng/mL in plasma (EDTA), and 500.03 ng/mL in plasma (heparin).

Serum diluted to 1 : 400 from ten individual healthy human female donors was measured in duplicate.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Ficolin-2 concentration was determined to be 403.80 ng/mL with a range of 138.63 – 755.53 ng/mL.

Product details

Human Ficolin-2 ELISA kit (ab275554) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Ficolin-2 protein in serum, plasma and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Ficolin-2 with 8.35 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less

-High sensitivity, specificity and reproducibility from superior antibodies

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Ficolin-2, initially identified as L-ficolin, in human is encoded by the FCN2 gene. Ficolin-2 belongs to the ficolin family of proteins and is characterized by the presence of a leader peptide, a short N-terminal segment, followed by a collagen-like region, and a C-terminal fibrinogen-like domain. Ficolin-2 is predominantly expressed in the liver. Studies have reported it to have roles in carbohydrate binding and opsonic activities. Serum concentration of ficolin-2 varies considerably in healthy individuals. FCN2 promoter polymorphisms were associated with marked changes in the ficolin-2 serum concentration.

Calcium-dependent lectin, which acts as a pattern recognition receptor that initiates the lectin pathway of the complement system, a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed : 10679061, PubMed : 14707097, PubMed : 15804047, PubMed : 17215869, PubMed : 25344472, PubMed : 8576206). Specifically recognizes and binds carbohydrates on the pathogen surface, activating the MASP1 serine protease and initiating the proteolytic cascade of the lectin complement pathway (PubMed : 14707097). Specifically binds N-Acetylglucosamine (GlcNAc), as well as phosphocholine and lipoteichoic acid moieties on the surface of pathogens (PubMed : 14707097, PubMed : 17215869, PubMed : 25344472, PubMed : 8576206). Enhances phagocytosis of S.typhimurium by neutrophils, suggesting an opsonic effect (PubMed : 8576206).
