The Human Ficolin-3 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213779) is designed for the quantitative measurement of Human Ficolin-3 in cell culture supernatants, serum and plasma (heparin).
The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Ficolin-3 has been pre-coated onto 96-well plates. Standards (Expression system for standard: NSO; Immunogen sequence: Q24-R299) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Ficolin-3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Ficolin-3 amount of sample captured in plate.
Ficolin-3, also known as HAKA1, is a protein that in humans is encoded by the FCN3 gene. It is mapped to 1p36.11. The protein can activate the complement pathway in association with MASPs and sMAP, thereby aiding in host defense through the activation of the lectin pathway. Ficolin-3 is expressed as a 35-kD protein that reacts with systemic lupus erythematosus sera. Ficolin-3 does not bind fibronectin (FN1), elastin (ELN), or zymosan, and the lectin activity of the Ficolin-3 is calcium independent. Homozygosity for the variant in the FCN3 gene results in a recessive complement deficiency syndrome.
