The Human Flt3 ligand / Flt3L Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213780) is designed for the quantitative detection of Human Flt3 ligand / Flt3L in cell culture supernatants, serum and plasma (heparin, EDTA, citrate).
The ELISA kit is based on standard sandwich enzyme-linked immunosorbent assay technology. A monoclonal antibody from mouse specific for Flt3 ligand / Flt3L has been pre-coated onto 96-well plates. Standards (Expression system for standard: NS0; Immunogen sequence: T27-P185) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Flt3 ligand / Flt3L is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Flt3 ligand / Flt3L amount of sample captured in plate.
Fms-related tyrosine kinase 3 ligand (FLT3LG), also known as Flt3 ligand / Flt3L or FL, is a protein which in humans is encoded by the FLT3LG gene. It is mapped to 19q13.3. Flt3 ligand / Flt3L is a hematopoietic four helical bundle cytokine. It is structurally homologous to stem cell factor (SCF) and colony stimulating factor 1 (CSF-1). In synergy with other growth factors, Flt3 ligand / Flt3L stimulates the proliferation and differentiation of various blood cell progenitors. Besides that, Flt3 ligand / Flt3L can control the development of Dendritic cells (DCs) and is particularly important for plasmacytoid DCs and CD8-positive classical DCs and their CD103-positive tissue counterparts. Flt3 ligand / Flt3L also can enhance the response of stem and primitive progenitor cells to other growth factors to generate all myeloid lineages except erythroid cells.

Biological function summary
The Flt3 ligand plays an essential role in the immune system. It is a monomeric cytokine that induces proliferation of early lineage cells within the hematopoietic system. Flt3L does not function as part of a complex but acts independently to stimulate the proliferation differentiation and survival of hematopoietic cells. By interacting directly with its receptor it significantly influences the innate and adaptive immune systems through the maturation of dendritic and B cells.
