1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

Follistatin is a single-chain polypeptide consisting of about 344 amino acids. The follistatin protein is encoded by a relatively small gene size of 6 kb and 6 exons with alternative splice variants and post translational modifications to give a variety of molecular weights between 31 – 39 kDa. Follistatin forms a high affinity complex with activin sequestering it from its receptor thus having an inhibitory effect. Follistatin acts to promote muscle growth in mammals by acting as an antagonist to the protein myostatin. Myostatin acts as an inhibitor to muscle growth.

Biological function summary
Follistatin modulates multiple processes such as cell growth differentiation and reproductive physiology. It acts as a binding protein for activin neutralizing its activity. By preventing activin from engaging with its receptor complexes follistatin influences developmental and regenerative processes. Despite not being a part of a transmembrane complex its interaction with activin impacts extracellular interactions.
Pathways
Follistatin plays a significant role in the regulation of the TGF-? signaling pathway and the muscle growth pathway. In the TGF-? pathway it counteracts the signaling of activin which influences cellular homeostasis and inflammatory responses. In the muscle growth pathway follistatin interferes with myostatin another member of the TGF-? family promoting muscle growth. This cross-talk with proteins such as myostatin demonstrates follistatin's influence in muscle regeneration and development.
