Interpolated concentrations of human FOXP3 in FOXP3 overexpressing HEK293 cell extract.
Interpolated concentration of overexpressed FOXP3 was measured in duplicate at different sample concentrations. Undiluted samples are 20 μg/mL FOXP3 overexpressing HEK293 cell extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Interpolated concentrations of FOXP3 in human FOXP3 overexpressing HEK293 cell extract.
The concentration of FOXP3 was measured in human FOXP3 overexpressing HEK293 cell extract and in empty plasmid transfected HEK293 cell extract, based on a 20 μg/mL extract load. The mean concentration was determined to be 24.73 ng/mL in FOXP3 overexpressing HEK293 cell extract and undetectable in empty plasmid transfected HEK293 cell extract.

Human FOXP3 ELISA Kit (ab289644) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human FOXP3 in cell extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human FOXP3 with 0.325 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
This transcription factor operates by regulating the expression of a variety of genes involved in Treg cell development and function. FOXP3 can interact with other proteins to form complexes that modulate gene expression. Its role is essential in enabling Tregs to suppress immune responses a process necessary for preventing autoimmunity. The interaction extends to other key regulators within Tregs such as the protein complex 221D which may have overlapping functional responsibilities. 'FOXP3 236a/E7' reflects its genetic variants detectable in different studies providing insights into its biological functions.
Pathways
FOXP3 is a significant player within the immune regulation pathway. It integrates into the TGF-beta signaling pathway in which it controls the expression of genes important for Treg functions. The protein recruits other transcription factors like NF-kB coordinating intricate network regulations within immune cells. FOXP3's activity orchestrates the balance between immune activation and suppression ensuring a well-functioning immune response that prevents overreactions leading to autoimmune conditions.
