The Human Galectin-4 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213785) is designed for the quantitative measurement of Human Galectin-4 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA).
The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Galectin-4 has been pre-coated onto 96-well plates. Standards (Expression system for standard: E.coli; Immunogen sequence: A2-I323) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Galectin-4 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human Galectin-4 amount of sample captured in plate.
Galectin-4, also known as LGALS4 or GAL4, is a protein that in humans is encoded by the LGALS4 gene. It is mapped to 19q13.2. Galectins are a family of animal lectins defined by an affinity for beta galactoside-containing saccharides and by shared sequence elements. Galectin-4 is expressed as a 38-kD protein in the human colon adenocarcinoma T84 cell line. Galectin-4 is an S-type lectin that is strongly under expressed in colorectal cancer, and this gene plays a role in cell adhesion. It has been found that decreased Galectin-4 expression occurs early in colorectal carcinogenesis.

Biological function summary
The GAL4 transcription factor functions as a major regulator of galactose-responsive genes. It acts as part of a complex binding with its DNA target sites to initiate transcription of GAL genes like GAL1 GAL7 and GAL10. Pathways
GAL4 is intimately involved in the GAL gene regulatory pathway that manages the cellular response to galactose. In this pathway proteins such as GAL80 inhibit GAL4 function under non-inducing conditions. However in the presence of galactose GAL3 binds and neutralizes GAL80 liberating GAL4 to activate gene expression. This system exemplifies a dynamic interaction in yeast allowing precise gene regulation in accordance with sugar availability.
