Supplementary information

Galectin 8 also known as LGALS8 is a member of the galectin family of proteins. It has a molecular mass of approximately 35 kDa and is found both in the cytoplasm and on cell surfaces across various tissues. Mechanically Galectin 8 binds to β-galactosides and displays dual carbohydrate-recognition domains allowing it to have diverse biological interactions. The widespread expression of Galectin 8 suggests its role in many different biological contexts serving as a critical participant in cellular growth adhesion and signaling processes.
Biological function summary

This protein contributes to various cellular processes including cell adhesion immune response and apoptosis regulation. Galectin 8 engages with cell adhesion molecules and receptors on the cell surface influencing cell-cell and cell-matrix interactions. It is not known to form a stable complex with other proteins but interacts with many proteins transiently to mediate its effects. The interactions facilitate cellular communication and migration which are processes important in both normal cellular function and disease states.

Pathways

Multiple cellular mechanisms involve Galectin 8 including the Wnt signaling and autophagy pathways. Within the Wnt signaling pathway Galectin 8 modulates cell proliferation and differentiation interacting with proteins like β-catenin and influencing gene expression. In autophagy it functions by interacting with the mammalian Target of Rapamycin (mTOR) signaling pathway where it helps to regulate cellular responses to nutrient availability and stress. These pathways illustrate the role of Galectin 8 in maintaining cellular homeostasis and responding to extracellular signals.

Evidence links Galectin 8 to autoimmune diseases and cancer. Its involvement in immune response modulation connects it to autoimmune disorders where abnormal expression may influence the onset or progression of conditions like rheumatoid arthritis. Moreover Galectin 8's role in cell adhesion and migration associates it with cancer progression particularly in tumor metastasis. Proteins such as integrins and matrix metalloproteinases collaborate with Galectin 8 in cancer highlighting the protein's broader impact on disease mechanisms.

Target data

Beta-galactoside-binding lectin that acts as a sensor of membrane damage caused by infection and restricts the proliferation of infecting pathogens by targeting them for autophagy (PubMed : 22246324, PubMed : 28077878). Detects membrane rupture by binding beta-galactoside ligands located on the lumenal side of the endosome membrane; these ligands becoming exposed to the cytoplasm following rupture (PubMed : 22246324, PubMed : 28077878). Restricts infection by initiating autophagy via interaction with CALCOCO2/NDP52 (PubMed : 22246324, PubMed : 28077878). Required to restrict infection of bacterial invasion such as S.typhimurium (PubMed : 22246324). Also required to restrict infection of Picornaviridae viruses (PubMed : 28077878). Has a marked preference for 3'-O-sialylated and 3'-O-sulfated glycans (PubMed : 21288902).
