Human GALNT3 ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human GALNT3 in serum, plasma and cell culture supernatants. This assay employs an antibody specific for human GALNT3 coated on a 96-well plate.

GALNT3 also known as polypeptide N-acetylgalactosaminyltransferase 3 is an enzyme that plays an important role in the O-glycosylation process. It transfers N-acetylgalactosamine (GalNAc) to serine and threonine residues of proteins. This enzyme belongs to the glycosyltransferase family and has a molecular mass of approximately 70 kDa. GALNT3 is mainly expressed in tissues such as liver kidney small intestine and bone.
Biological function summary

GALNT3 is responsible for initiating O-linked glycosylation of proteins affecting their stability function and localization. It acts alone and not as part of a larger protein complex. Through its activity GALNT3 modulates various proteins involved in cellular functions such as signal transduction and cell-cell interaction. This glycosylation process helps in the correct folding of proteins and protecting them from proteolytic degradation.

Pathways

The role of GALNT3 places it prominently in the glycosylation pathway where it influences the modification of proteins involved in the signaling cascade. One important pathway involving GALNT3 is the regulation of mineralization controlled by fibroblast growth factor 23 (FGF23). GALNT3 glycosylates and activates FGF23 highlighting its relationship to other proteins within the signaling axis including the fibroblast growth factor receptors (FGFRs).

Alterations in GALNT3 are associated with familial tumoral calcinosis a disorder characterized by abnormal calcium and phosphate metabolism. Mutations in GALNT3 disrupt its function leading to improper glycosylation and degradation of FGF23 resulting in elevated levels of phosphate. The connection between GALNT3 and FGF23 is central in understanding the pathogenesis of this condition as well as other metabolic bone diseases related to phosphate homeostasis.
