Sandwich ELISA - Human gamma-synuclein ELISA Kit (AB224884)
Example of human gamma-synuclein standard curve in Sample Diluent NS.
The gamma-synuclein standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Sandwich ELISA - Human gamma-synuclein ELISA Kit (AB224884)
Urine from pooled normal human and four individual bladder cancer patients (male : n=3; female : n=1) human donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean gamma-synuclein concentration was determined to be 569.88 pg/mL with a range of 40.46 – 2021.56 pg/ml. The native concentration of gamma-synuclein was below the level of detection in urine from healthy human donors.

Sandwich ELISA - Human gamma-synuclein ELISA Kit (AB224884)
Interpolated concentrations of native gamma-synuclein in human serum and plasma samples.
The concentrations of gamma-synuclein were measured in duplicate, interpolated from the gamma-synuclein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean gamma-synuclein concentration was determined to be 126.89 pg/mL in serum, 114.40 pg/mL in plasma (citrate), 78.63 pg/mL in plasma (EDTA) and 173.43 pg/ml in plasma (heparin).

Sandwich ELISA - Human gamma-synuclein ELISA Kit (AB224884)
Interpolated concentrations of native gamma-synuclein in human urine (from a bladder cancer patient) and cell culture extract based on a 5 μg/mL load.
The concentrations of gamma-synuclein were measured in duplicate and interpolated from the gamma-synuclein standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean gamma-synuclein concentration was determined to be 2193.46 pg/mL in bladder cancer urine, 316.85 pg/ml in HT-29 cell extract.

Product details
Human gamma-synuclein ELISA Kit (ab224884) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of gamma-synuclein protein in cit plasma, edta plasma, hep plasma, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human gamma-synuclein with 5.15 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Human gamma-synuclein plays a role in neurofilament network integrity. May be involved in modulating axonal architecture during development and in the adult. In vitro, increases the susceptibility of neurofilament-H to calcium-dependent proteases (By similarity). May also function in modulating the keratin network in skin. Activates the MAPK and Elk-1 signal transduction pathway.

Biological function summary
Gamma synuclein plays a role in modulating neuronal plasticity and may participate in synaptic regulation. It is not known to be a part of a larger protein complex but it interacts with various other proteins influencing neuron differentiation. Although its precise biological functions are not fully characterized studies suggest that gamma synuclein could impact cellular processes like cytoskeletal dynamics and membrane trafficking.
Pathways
The protein gamma synuclein is involved in cellular stress response and neurodegeneration pathways. It is closely related to proteins like alpha synuclein with which it shares structural and functional features. Pathways involving these proteins can influence several processes such as axonal transport and neurotransmitter release. Dysregulation in these pathways often leads to disruptions in cellular homeostasis.
