Product details
Human Gas 1 ELISA Kit is designed for the quantitative determination of Gas 1 in cell culture supernatants, plasma and serum samples.
This assay employs an antibody specific for human Gas 1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Gas 1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human Gas 1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Gas 1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
Gas1 acts as a negative regulator of cell proliferation particularly during developmental processes. It is not part of a complex but serves as a standalone inhibitor that influences cell growth. The protein interacts with components like Hedgehog signaling which leads to its involvement in developmental signaling pathways. Gas1's role in growth arrest demonstrates its importance in controlling the balance of cellular differentiation and proliferation.
Pathways
Gas1 significantly influences the Hedgehog signaling pathway and the G1 phase of the cell cycle. In the Hedgehog pathway Gas1 interacts with other key proteins like Patched and Smoothened regulating the pathway's activity which is essential for embryonic development and tissue patterning. Its interaction with these proteins highlights its regulatory function in signal transduction influencing how cells respond to growth cues.
