Interpolated concentration of native GATA4 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : SH-SY5Y extract 250 µg/mL, and human heart extract 500 µg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Human GATA4 SimpleStep ELISA^® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of GATA4 protein in human cell and tissue extracts. Quantitate Human GATA4 with 4.02 pg/ml sensitivity.

SimpleStep ELISA^® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA^® protocol summary in the image section for further details. Our SimpleStep ELISA^® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples

A 384-well SimpleStep ELISA^® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA^® kits.

GATA4 also known as GATA-binding protein 4 is a transcription factor that plays a significant role in gene expression regulation. It weighs approximately 44 kDa. This protein expresses mainly in cells of the heart gonads and liver. GATA4 functions through binding to the GATA sequence on DNA regulating various genes. The protein is well known for its involvement in cardiac development and function.

Transcriptional activator that binds to the consensus sequence 5'-AGATAG-3' and plays a key role in cardiac development and function (PubMed : 24000169, PubMed : 27984724, PubMed : 35182466). In cooperation with TBX5, it binds to cardiac super-enhancers and promotes cardiomyocyte gene expression, while it down-regulates endocardial and endothelial gene expression (PubMed : 27984724). Involved in bone morphogenetic protein (BMP)-mediated induction of cardiac-specific gene expression.
