Interpolated concentrations of native GFAP in Human brain extract based on a 100 ng/mL extract load.
The concentrations of GFAP were measured in duplicate and interpolated from the GFAP standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean GFAP concentration was determined to be 46.27 ng/mL per 100 ng/ml extract load.

How the assay works
Human GFAP SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Glial fibrillary acidic protein (GFAP) is a class-III intermediate filament, cell-specific marker that during development of the central nervous system, distinguishes astrocytes from other glial cells. Protein expression is also found in numerous cell types including astrocytes, ependymal cells, osteocytes, keratinocytes and chondrocytes. GFAP has also been found to be expressed in rat glomeruli, peritubular fibroblasts, stellate cells of the pancreas and liver tissue as well as Leydig cells of the testis in both hamsters and humans.
