Interpolated concentrations of spiked GITR in human serum, plasma and cell culture supernatant samples.
The concentrations of GITR were measured in duplicates, interpolated from the GITR standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (EDTA) 25%, plasma (citrate) 25%, plasma (heparin) 25% and cell culture media 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Interpolated concentrations of native GITR in human MCF7 cell extract based on a 1,000 μg/mL extract load.
The concentrations of GITR were measured in duplicate and interpolated from the GITR standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean GITR concentration was determined to be 757 pg/mL in MCF7 cell extract.

Human GITR ELISA Kit (ab234571) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of GITR protein in cell culture extracts, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human GITR with 14 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies

Biological function summary
GITR acts as a costimulatory molecule that enhances T cell activation and proliferation. It does not function as part of a complex but interacts directly with ligand GITRL triggering downstream signaling cascades. This interaction is key in the modulation of immune responses notably in enhancing the activity of effector T cells and regulating Treg functions. Activation of GITR can reduce Treg-mediated suppression leading to enhanced immune responses.
Pathways
The mechanistic role of GITR involves NF-kB and MAPK signaling pathways. Through these GITR impacts immune responses by stimulating the production of cytokines and promoting cell survival and proliferation. GITR signaling intersects with pathways involving proteins such as NF-kB further integrating with the immune system's regulation crescendo.
