Interpolated concentrations of native Glycogen Synthase 1 in mouse NIH3T3 extract and rat PC12 extract.
The concentrations of Glycogen Synthase 1 were measured in duplicate and interpolated from the Glycogen Synthase 1 standard curve and corrected for sample dilution and extract load. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Glycogen Synthase 1 concentration was determined to be 5.7 pg/μg NIH3T3 extract and 595 pg/μg PC12 extract

How the assay works
Human Glycogen Synthase 1 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Glycogen Synthase 1 (also known as Glycogen [starch] synthase, muscle) is a glycosyltransferase enzyme involved in converting glucose to glycogen. Mutations in Glycogen Synthase 1 are associated with glycogen storage disease. Mouse and rat Glycogen Synthase 1 have 97% protein sequence identity to human Glycogen Synthase 1.

Biological function summary
This enzyme plays a critical role in the energy storage mechanism of muscle tissues. GYS1 functions as a part of the glycogen synthase complex interacting with various glycogen-associated proteins to modulate its activity. These interactions control the enzyme's affinity for its substrate and its susceptibility to regulatory signals like phosphorylation affecting glycogen synthesis efficiency. Through its activity GYS1 ensures a readily available reserve of glucose in muscle tissues for rapid energy production when required.
Pathways
Glycogen synthase 1 is a central component of the glycogen metabolism pathway. It acts alongside other enzymes such as glycogen phosphorylase which catalyzes glycogen breakdown. The balance between GYS1 and glycogen phosphorylase determines the rate of glycogen turnover within cells. GYS1 is further regulated by the insulin signaling pathway where insulin promotes glycogen synthesis by altering GYS1 activity alongside proteins like AKT influencing glucose homeostasis and energy management in tissues.
