Abcam's GM-CSF Receptor alpha (CD116) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of GM-CSF R alpha in serum, plasma, and cell culture supernatants.

A monoclonal antibody specific for GM-CSF R alpha has been coated onto the wells of the microtiter strips provided. Samples, including standards of known GM-CSF R alpha concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for GM-CSF R alpha are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of GM-CSF R alpha present in the samples.

This kit will recognize both endogenous and recombinant Human GM-CSF R alpha.

Activity summary

The GM-CSF receptor alpha also known as CD116 is a protein that plays role in the immune response. It is a component of the GM-CSF receptor complex and has an approximate molecular weight of 74 kDa. This receptor is found on the surface of various cell types like monocytes macrophages and neutrophils. It binds specifically to granulocyte-macrophage colony-stimulating factor (GM-CSF) which leads to the activation of downstream signaling pathways important for cell survival and proliferation.

Biological function summary

GM-CSF receptor alpha binds GM-CSF with high affinity and is part of a receptor complex that includes beta common chain (βc). The receptor complex initiates signaling cascades that modulate immune functions such as differentiation of myeloid cells and macrophage activation. These interactions support various immune processes like inflammation and pathogen defense. GM-CSF receptor alpha is essential to ensure proper cellular responses to environmental cues.

Pathways

GM-CSF receptor alpha is involved in the JAK-STAT and MAPK pathways. Upon ligand binding it activates JAK2 which in turn triggers STAT5 phosphorylation — important for transcription of immune-related genes. Simultaneously it initiates the MAPK (mitogen-activated protein kinase) pathway further promoting cell differentiation and proliferation. Each of these signaling events connect GM-CSF receptor alpha to other proteins like JAK2 and STAT5 which collaborate to sustain proper immune activity.

Associated diseases and disorders

GM-CSF receptor alpha is linked to pulmonary alveolar proteinosis and rheumatoid arthritis. In pulmonary alveolar proteinosis dysregulation of GM-CSF receptor signaling leads to impaired surfactant clearance resulting in respiratory complications. In rheumatoid arthritis increased GM-CSF signaling contributes to inflammation and joint damage. Proteins like βc and GM-CSF interact with GM-CSF receptor alpha amplifying the pathological signals in these conditions highlighting its potential as a therapeutic target.
