Interpolated concentrations of human HAI-1 in PBMC supernatant stimulated for 46 hours with 1.5% PHA-M. Interpolated concentration of native HAI-1 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : stimulated PBMC supernatant (100%). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Interpolated concentrations of HAI-1 in normal human serum donors. Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +- SD, n=2). The mean HAI-1 concentration was determined to be 93.19 ng/mL with a range of 54.39 - 161.95 ng/mL.

Human HAI-1 ELISA Kit (ab288176) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human HAI-1 protein in cell culture supernatant, plasma and serum samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human HAI-1 with 0.29pg/mL sensitivity
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
HAI-1 performs a regulatory role by inhibiting proteases such as matriptase and hepsin which are critical for many cellular functions. As part of a complex HAI-1 binds to these proteases controlling their activity to prevent excessive proteolysis which can damage tissues. This regulatory function is key in maintaining epithelial integrity and homeostasis impacting processes like cell growth and migration. Anti-HAI studies show that disrupting this balance can lead to a cascade of biological alterations.
Pathways
Several cellular processes heavily depend on the balance maintained by HAI-1 in protease activity. One significant pathway involves the HGF/c-Met signaling cascade which impacts cell motility and proliferation. By inhibiting matriptase HAI-1 indirectly affects this pathway regulating cellular responses. Additionally HAI-1 also relates to epithelial-mesenchymal transition (EMT) through controlling proteolytic activity which can influence how cells migrate and differentiate in development or healing.
