Abcam's HB EGF Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human HB EGF in plasma and cell culture supernatants. (Human HB EGF concentration is pretty low in normal plasma, it may not be detected in this assay).

This assay employs an antibody specific for Human HB EGF coated on a 96-well plate. Standards and samples are pipetted into the wells and HB EGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human HB EGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of HB EGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

We have not been able to detect the endogenous Human HB-EGF in normal serum with ab100531, only in serum spiked with Human HB-EGF.

Optimization may be required with urine samples.

The standard is affinity purified.

HB-EGF also known as heparin-binding EGF-like growth factor is a member of the EGF family with an approximate molecular mass of 19-23 kDa. It is a membrane-anchored growth factor that can be cleaved to release a soluble form. HB-EGF expresses in a wide range of tissues including the heart kidneys lungs and the brain. As a growth factor its primary mechanical function involves binding to EGFR (epidermal growth factor receptor) and ERBB4 receptors stimulating cellular proliferation and survival.
Biological function summary

The actions of HB-EGF extend beyond simple growth factor roles. It participates as a modulator in wound healing processes cardiac development and neural tissue maintenance. The protein often functions as part of a signaling complex that involves other growth factors and receptors. Its engagement in biological processes links directly to modifying cellular behavior such as enhancing cell migration and inducing tissue repair mechanisms.

Pathways

Growth factor that mediates its effects via EGFR, ERBB2 and ERBB4. Required for normal cardiac valve formation and normal heart function. Promotes smooth muscle cell proliferation. May be involved in macrophage-mediated cellular proliferation. It is mitogenic for fibroblasts, but not endothelial cells. It is able to bind EGF receptor/EGFR with higher affinity than EGF itself and is a far more potent mitogen for smooth muscle cells than EGF. Also acts as a diphtheria toxin receptor.
