Abcam's HCC-1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay designed for the quantitative measurement of Human HCC-1 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human HCC-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and HCC-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human HCC-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimization may be required with urine samples.

Biological function summary
HCC-1 affects immune response by interacting with chemokine receptors. It binds particularly to CCR1 and CCR5 receptors on the cell surface. Though HCC-1 is not usually part of a larger protein complex it functions closely with other chemokines in modulating leukocyte behavior. These interactions highlight its role in immune surveillance and inflammatory response demonstrating its significance in orchestrating an effective immune defense.
Pathways
HCC-1 is actively involved in chemokine signaling pathways and has a role in inflammation-related processes. Its interaction with CCR1 and CCR5 connects it directly to the MAPK signaling pathway impacting cell proliferation and survival. HCC-1 similar to proteins such as MCP-1 and RANTES plays a part in guiding immune cells during inflammation therefore contributing to the overall coordination of the immune response.
