Product details
Human hCG ELISA Kit ab100533 is a sandwich ELISA to measure Human hCG in serum, plasma, cell culture supernatant with a sensitivity of 50 pg/ml.
How the assay works
This assay employs an antibody specific for Human hCG coated on a 96-well plate. Standards and samples are pipetted into the wells and hCG present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human hCG antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of hCG bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

1. Add standard or sample to each well used. Incubate at room temperature
2. Add prepared biotin antibody to each well. Incubate at room temperature.
3. Add prepared Streptavidin solution. Incubate at room temperature
4. Add TMB One-Step Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.

Biological function summary
HCG regulates several functions related to reproduction. It supports the corpus luteum in ovaries to secrete progesterone during early pregnancy contributing to the uterine lining's maintenance.
