Sandwich ELISA - Human Heme Oxygenase 1 ELISA Kit (AB207621)
Interpolated concentrations of native Heme Oxygenase 1 in human liver homogenate extract and A549 cell extract based on a 15.6 μg/mL liver homogenate extract and 5 μg/mL A549 cell extract load.
The concentrations of Heme Oxygenase 1 were measured in duplicate and interpolated from the Heme Oxygenase 1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Heme Oxygenase 1 concentration was determined to be 3.30 ng/mL in liver homogenate extract and 1.60 ng/mL in A549 cell extract.

Sandwich ELISA - Human Heme Oxygenase 1 ELISA Kit (AB207621)
Interpolated concentrations of native Heme Oxygenase 1 in human serum and plasma (EDTA) samples.
The concentrations of Heme Oxygenase 1 were measured in duplicate, interpolated from the Heme Oxygenase 1 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Heme Oxygenase 1 concentration was determined to be 1.37 ng/mL in serum and 2.12 ng/mL in plasma (EDTA).

Product details
Human Heme Oxygenase 1 ELISA Kit ab207621 is a rapid single-wash 90-min Sandwich ELISA to measure Human Heme Oxygenase 1 in cell culture extracts, cell culture supernatant, citrate plasma, EDTA plasma, heparin plasma, serum, tissue extracts. This SimpleStep sensitivity is 7.9 pg/mL.
How the assay works
Human Heme Oxygenase 1 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

We offer the antibody pair used in this kit in a BSA and Azide-free format, ready for conjugation:
- Anti-Heme Oxygenase 1 antibody [EPR18689-5] - BSA and Azide free (Capture) ab242982
- Anti-Heme Oxygenase 1 antibody [EPR18689-126] - BSA and Azide free (Detector) ab242982
Heme oxygenase 1 (HO 1) is an enzyme that functions in heme catabolism. The activity of heme oxygenase 1 is to cleave the heme ring to form biliverdin. There are two isozymes of heme oxygenase that have 56% amino acid sequence identity: inducible heme oxygenase 1 and constitutively expressed heme oxygenase 2.

Biological function summary
The function of HO-1 involves important antioxidant defense mechanisms. The enzyme contributes to the cytoprotective effects by moderating oxidative stress and inflammation through its enzymatic activity. HO-1 often forms part of a larger regulatory complex where its activity regulates other cellular processes. Its heme byproducts biliverdin and bilirubin possess strong antioxidant properties adding to the cell's capacity to neutralize harmful oxidative effects.
Pathways
HO-1 operates in heme degradation and the antioxidative response pathway. It closely interacts with ferritin in its role of managing intracellular iron levels. As HO-1 breaks down heme the free iron released is sequestered by ferritin preventing cellular iron overload. HO-1 also plays a part in the signaling pathway involved in maintaining vascular homeostasis where it may modulate the activity of nitric oxide synthase through its byproducts.
