Heme oxygenase 1. Catalyzes the oxidative cleavage of heme at the alpha-methene bridge carbon, released as carbon monoxide (CO), to generate biliverdin IXalpha, while releasing the central heme iron chelate as ferrous iron (PubMed:11121422, PubMed:19556236, PubMed:7703255). Affords protection against programmed cell death and this cytoprotective effect relies on its ability to catabolize free heme and prevent it from sensitizing cells to undergo apoptosis (PubMed:20055707). Heme oxygenase 1. (Microbial infection) During SARS-COV-2 infection, promotes SARS-CoV-2 ORF3A-mediated autophagy but is unlikely to be required for ORF3A-mediated induction of reticulophagy. Heme oxygenase 1 soluble form. Human Heme Oxygenase 1 ELISA Kit (ab207621) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Heme Oxygenase 1 protein in cell culture extracts, cell culture supernatant, serum, tissue extracts, cit plasma, edta plasma, and hep plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Heme Oxygenase 1 with 7.9 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less

ASSAY SPECIFICITY

This kit recognizes both native and recombinant human Heme Oxygenase 1 protein in serum, plasma, cell culture supernatant, and cell and tissue extract samples.

SPECIES REACTIVITY

This kit recognizes human Heme Oxygenase 1 protein.
Other species reactivity was determined by measuring 50% (dilution) serum samples of various species, interpolating the protein concentrations from the human standard curve, and expressing the interpolated concentrations as a percentage of the protein concentration in human serum assayed at the same dilution. Reactivity < 3% was determined for the following species: Mouse , Rat , Hamster, Guinea Pig , Dog, Goat, Pig, Cow

Heme oxygenase 1 (HO 1) is an enzyme that functions in heme catabolism. The activity of heme oxygenase 1 is to cleave the heme ring to form biliverdin. There are two isozymes of heme oxygenase that have 56% amino acid sequence identity: inducible heme oxygenase 1 and constitutively expressed heme oxygenase 2.

Activity summary

Heme oxygenase 1 (HO-1) also known as heme oxygenase-1 or oxygenase catalyzes the degradation of heme into biliverdin carbon monoxide and free iron. It has been characterized with a mass of approximately 32 kDa. HO-1 is widely expressed in tissues with high metabolic activity such as liver spleen bone marrow and brain. This enzyme plays key roles in cellular response to oxidative stress and is responsive to various stress-inducing stimuli.

Biological function summary

The function of HO-1 involves important antioxidant defense mechanisms. The enzyme contributes to the cytoprotective effects by moderating oxidative stress and inflammation through its enzymatic activity. HO-1 often forms part of a larger regulatory complex where its activity regulates other cellular processes. Its heme byproducts biliverdin and bilirubin possess strong antioxidant properties adding to the cell's capacity to neutralize harmful oxidative effects.

Pathways

HO-1 operates in heme degradation and the antioxidative response pathway. It closely interacts with ferritin in its role of managing intracellular iron levels. As HO-1 breaks down heme the free iron released is sequestered by ferritin preventing cellular iron overload. HO-1 also plays a part in the signaling pathway involved in maintaining vascular homeostasis where it may modulate the activity of nitric oxide synthase through its byproducts.

Associated diseases and disorders

Dysregulation of HO-1 levels connects to conditions such as cardiovascular diseases and certain neurodegenerative disorders. In cardiovascular diseases especially atherosclerosis the improper balance of heme oxygenase-1 activity can influence the severity of oxidative damage. Furthermore HO-1 relates to the pathogenesis of Alzheimer's disease where oxidative stress plays a significant role and where its interaction with proteins like amyloid-beta if disrupted may worsen disease pathology.

    The concentrations of Heme Oxygenase 1 were measured in duplicate, interpolated from the Heme Oxygenase 1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Heme Oxygenase 1 concentration was determined to be 1.37 ng/mL in serum and 2.12 ng/mL in plasma (EDTA).
