How the assay works

Human Heme Oxygenase 1 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Heme oxygenase 1 (HO 1) is an enzyme that functions in heme catabolism. The activity of heme oxygenase 1 is to cleave the heme ring to form biliverdin. There are two isozymes of heme oxygenase that have 56% amino acid sequence identity: inducible heme oxygenase 1 and constitutively expressed heme oxygenase 2.

Heme oxygenase 1 (HO-1) also known as heme oxygenase-1 or oxygenase catalyzes the degradation of heme into biliverdin carbon monoxide and free iron. It has been characterized with a mass of approximately 32 kDa. HO-1 is widely expressed in tissues with high metabolic activity such as liver spleen bone marrow and brain. This enzyme plays key roles in cellular response to oxidative stress and is responsive to various stress-inducing stimuli.
Biological function summary

The function of HO-1 involves important antioxidant defense mechanisms. The enzyme contributes to the cytoprotective effects by moderating oxidative stress and inflammation through its enzymatic activity. HO-1 often forms part of a larger regulatory complex where its activity regulates other cellular processes. Its heme byproducts biliverdin and bilirubin possess strong antioxidant properties adding to the cell's capacity to neutralize harmful oxidative effects.

Pathways

HO-1 operates in heme degradation and the antioxidative response pathway. It closely interacts with ferritin in its role of managing intracellular iron levels. As HO-1 breaks down heme the free iron released is sequestered by ferritin preventing cellular iron overload. HO-1 also plays a part in the signaling pathway involved in maintaining vascular homeostasis where it may modulate the activity of nitric oxide synthase through its byproducts.

Dysregulation of HO-1 levels connects to conditions such as cardiovascular diseases and certain neurodegenerative disorders. In cardiovascular diseases especially atherosclerosis the improper balance of heme oxygenase-1 activity can influence the severity of oxidative damage. Furthermore HO-1 relates to the pathogenesis of Alzheimer's disease where oxidative stress plays a significant role and where its interaction with proteins like amyloid-beta if disrupted may worsen disease pathology.
