Human HGFA ELISA Kit is designed for the quantitative determination of HGFA in cell culture supernatants, plasma and serum samples.
This assay employs an antibody specific for human HGFA coated on a 96-well plate. Standards and samples are pipetted into the wells and HGFA present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human HGFA antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of HGFA bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The HGF activator also known as HGFA is a serine protease that plays a critical role in activating hepatocyte growth factor (HGF). HGFA is present in blood plasma as an inactive single chain precursor with a molecular mass of approximately 70 kDa. It undergoes proteolytic cleavage to become active. This activator is highly expressed in the liver but can also be found in other tissues including the lung kidney and placenta. HGFA is essential in processes that require remodeling or repair as it activates HGF by cleaving to expose its receptor binding site.
HGFA initiates important physiological processes by transforming inactive HGF into its active form. It is not part of a large complex but interacts closely with HGF and its receptor MET. Through these interactions HGFA plays a significant role in tissue regeneration cell growth and motility. The HGFA-mediated activation of HGF also contributes to embryonic development and wound healing by promoting cell migration and angiogenesis. This interaction highlights the importance of HGFA in development and repair processes within the body.
HGFA operates significantly within the HGF/MET signaling pathway. This pathway is essential for cellular mechanisms like mitogenesis morphogenesis and motility. Additionally the activator has an indirect connection to the PI3K/AKT pathway as HGF signaling can lead to downstream activation of AKT which influences cell survival and proliferation. HGFA’s influences extend through these pathways impacting MET as an important receptor protein and connecting to other actors within these pathways.
HGFA is linked notably to cancer and tissue fibrosis. The dysregulation of HGF/MET signaling with HGFA playing a central role is often seen in tumorigenesis particularly in cancers like hepatocellular carcinoma and gastric cancer. Overactive HGFA can lead to excessive activation of HGF which in turn stimulates uncontrolled cell proliferation through MET. Moreover the contribution of HGFA to wound healing processes can exacerbate conditions like liver fibrosis where HGF’s role in fibrogenesis is marked. Understanding the function of HGFA helps in elucidating its contribution to these diseases positioning it as a potential target for therapeutic intervention.
