Interpolated concentrations of native HLA-DR in human cell and tissue extract samples.
The concentrations of HLA-DR were measured in duplicates, interpolated from the HLA-DR standard curves corrected for sample dilution and extract load. Undiluted samples are as follows : Daudi cell extract 32.3 μg/mL, Raji cell extract 490 μg/mL, and human skin extract 500 μg/mL. The interpolated dilution factor and corrected values are plotted (mean +/- SD, n=2). The mean HLA-DR concentration was determined to be 52.0 pg/μg in Daudi Extract, 1.93 pg/μg in Raji Extract and 0.85 pg/μg in human skin homogenate extract.

Human HLA-DR ELISA Kit ab223593 is a rapid single-wash 90-min Sandwich ELISA to measure Human HLA-DR in cell culture extracts, tissue homogenate. This SimpleStep sensitivity is 5 pg/mL.
How the assay works
Human HLA-DR SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

HLA class II histocompatibility antigen, DR alpha chain (HLA-DR) is the alpha chain of a cell surface receptor heterodimer that binds peptides for presentation to CD4 T cells. The alpha chain is fairly constant between individuals and pairs with varying alleles of the beta chain. HLA-DR has been implicated in multiple auto-immune conditions, as well as graft vs host disease.
