Interpolated concentrations of native HNF4-alpha in human HepG2 extract and based on a 40 μg/mL extract load.

The concentrations of HNF4a were measured in duplicate and interpolated from the HNF4-alpha standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean HNF4-alpha concentration was determined to be 1346 μg/mL in HepG2 extract.

How the assay works

Human HNF-4-alpha SimpleStep ELISA^{^®}employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^{^®} plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. HNF-4-alpha also known as hepatocyte nuclear factor 4-alpha (HNF4A) is a nuclear protein that functions as a transcription factor. It plays an important role in regulating gene expression in the liver kidney and intestine. HNF-4-alpha has a molecular mass of about 54 kDa and is expressed in tissues where it maintains cellular differentiation and metabolic homeostasis. In cellular assays such as ELISA and CHIP detection of HNF-4-alpha provides insights into its expression levels and activity in various biological contexts.
