Interpolated concentrations of native ICAM3 in human serum, plasma (EDTA), plasma (citrate), and plasma (heparin).
The concentrations of ICAM3 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5%, plasma (EDTA) 12.5%, plasma (citrate) 12.5%, and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 124.4 ng/mL in serum, 87.3 ng/mL in plasma (EDTA), 96.7 ng/mL in and plasma (citrate), 105 ng/mL in plasma (heparin).

Serum from eight individual healthy ICAM3 female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ICAM3 concentration was determined to be 87.3 ng/mL with a range of 51 – 128 ng/mL.

Human ICAM3 ELISA kit (ab275097) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ICAM3 protein in serum, plasma and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ICAM3 with 0.036 ng/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Intracellular adhesion molecule 3 (ICAM-3), also known as CD50, is a protein encoded by the gene ICAM3. ICAM3 belongs to the ICAM family of proteins, where all ICAM proteins are type I transmembrane glycoproteins, contain several immunoglobulin-like c2 domains, and bind to leukocyte adhesion protein (LFA-1). In addition, ICAM-3 is a ligand for integrin alpha-D/beta-2, where it contributes to apoptotic neutrophil phagocytosis by macrophages.

Biological function summary
ICAM-3 contributes to the immune system’s functionality by mediating intercellular adhesion and cellular signaling. It acts as a ligand for the integrin LFA-1 found on leukocytes which facilitates the initial immune response. ICAM-3 is not a part of a larger complex but works closely with LFA-1 in homing and recruitment of immune cells to inflammation sites. Its interaction with LFA-1 also supports the formation of the immunological synapse an important step in T-cell activation and immune signaling.

ICAM-3 contributes to both the immune response and leukocyte extravasation pathways. The interaction with LFA-1 places it within the context of immune cell signaling pathways impacting integrin-mediated signaling. Another pathway involving ICAM-3 is the cell adhesion pathway which is essential for maintaining proper immune surveillance and response. It collaborates with proteins like ICAM-1 in these pathways to ensure effective immune activity.
