Interpolated concentrations of native ICOS Ligand in human HEK293 overexpression extract based on a 5 μg/mL extract load.
The concentrations of ICOS Ligand were measured in duplicate and interpolated from the ICOS Ligand standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Human ICOS Ligand ELISA kit (ab272475) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human ICOS Ligand protein in cell extract and culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human ICOS Ligand with 21.8 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Inducible T Cell Co-Stimulator Ligand (ICOSLG), also known as CD275 and B7-H2, is a glycoprotein encoded by the gene ICOSLG. ICOS Ligand is a single pass type I membrane protein, which consists of a 238 -amino acid extracellular portion, a 21- amino acid transmembrane region, and a 25-amino acid cytoplasmic region. ICOS Ligand is constitutively expressed in many tissues and is enhanced by treatment with TNF in peripheral blood B-cells and monocytes. ICOS acts as a costimulatory signal for T-cell proliferation and cytokine secretion. ICOS Ligand induces B-Cell proliferation and differentiation into plasma cells.

Biological function summary
ICOSL contributes to the modulation of immune responses. By forming a complex with the active ICOS protein ICOSL is involved in the co-stimulation of T cells. This process is important for the development of T-cell-dependent immune responses and helps in promoting the survival and differentiation of T cells. Together the ICOS ligand and ICOS protein create a dynamic interaction that balances immune activation and tolerance which is essential for maintaining immune homeostasis.
Pathways
The ICOS-ICOSL interaction plays an important role in the T-cell receptor (TCR) signaling pathway. This pathway is fundamental for the activation of T cells and subsequent immune response. ICOSL also connects to the PI3K/Akt pathway where it impacts cell survival and proliferation. ICOS and the PI3K/Akt pathway work together to influence various aspects of immune regulation and adaptive immunity.
