Interpolated concentrations of human IDH1 in Raji cell extract.
Interpolated concentration of native IDH1 was measured in duplicate at different sample concentrations. Undiluted sample 200 μg/mL Raji cell extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent 1X Cell Extraction Buffer PTR.

Interpolated concentrations of IDH1 in human wild type and knockout cell line cell extracts.
The concentration of IDH1 was measured in human wild type and knockout Hela cell line cell extracts based on a 100 μg/mL extract load. The concentration at 100 μg/mL was determined to be 44.1 ng/mL in wild type extract and undetectable in knockout extract. The control and IDH1 knockout cell line extracts are available as ab260873.

Human IDH1 SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IDH1 protein in human serum, plasma, saliva and cell culture supernatant. Quantitate Human IDH1 with 0.072 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Biological function summary
Isocitrate dehydrogenase 1 plays an important role in cellular metabolism by helping to maintain the NADP+/NADPH balance important for redox reactions. This enzyme operates mainly as a homodimer meaning two identical subunits form a complex for functional activity. It contributes significantly to lipid and glucose metabolism due to its production of NADPH which the cells use for biosynthetic pathways and detoxification.
Pathways
The involvement of IDH1 in the citric acid cycle highlights its importance in energy production and metabolic regulation. This enzyme closely associates with other metabolic enzymes such as IDH2 another member of the isocitrate dehydrogenase family found in mitochondria suggesting a coordinated function between cytoplasmic and mitochondrial metabolism. Beyond its role in the citric acid cycle IDH1 also relates to the pentose phosphate pathway an alternative glucose metabolism route providing reducing equivalents and ribose 5-phosphate for nucleotide synthesis.
