Interpolated concentrations of native IGFBP1 in human milk and cell culture supernatant samples.

The concentrations of IGFBP1 were measured in duplicates, interpolated from the IGFBP1 standard curves and corrected for sample dilution. Undiluted samples are as follows : milk 5%, A549 supernatant 100%, and HepG2 supernatant 0.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 12 ng/ml in milk, 0.5 ng/ml in A549 supernatant, and 172 ng/mL in HepG2 supernatant.

Interpolated concentrations of native IGFBP1 in human HepG2, U-87 MG, and liver homogenate extract samples based on a 4 μg/mL, 500 μg/ml, and 500 μg/mL extract load respectively.

The concentrations of IGFBP1 were measured in duplicate and interpolated from the IGFBP1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP1 concentration was determined to be 580 pg/mL in HepG2 cell extract, 457 pg/ml in U-87 MG cell extract, and 739 pg/mL in liver tissue homogenate extract samples.

Human IGFBP1 ELISA Kit (ab233618) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IGFBP1 protein in cit plasma, edta plasma, hep plasma, milk, serum, tissue homogenate, urine, cell culture supernatant, and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IGFBP1 with 11.6 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Multifunctional protein that plays a critical role in regulating the availability of IGFs such as IGF1 and IGF2 to their receptors and thereby regulates IGF-mediated cellular processes including cell migration, proliferation, differentiation or apoptosis in a cell-type specific manner (PubMed : 11397844, PubMed : 15972819). Plays also a positive role in cell migration by interacting with integrin ITGA5 : ITGB1 through its RGD motif (PubMed : 7504269). Mechanistically, binding to integrins leads to activation of focal adhesion kinase/PTK2 and stimulation of the mitogen-activated protein kinase (MAPK) pathway (PubMed : 11397844). Regulates cardiomyocyte apoptosis by suppressing HIF-1alpha/HIF1A ubiquitination and subsequent degradation (By similarity).
