Multifunctional protein that plays a critical role in regulating the availability of IGFs such as IGF1 and IGF2 to their receptors and thereby regulates IGF-mediated cellular processes including proliferation, differentiation, and apoptosis in a cell-type specific manner (PubMed:10874028, PubMed:19556345). Also exhibits IGF-independent antiproliferative and apoptotic effects mediated by its receptor TMEM219/IGFBP-3R (PubMed:20353938). Inhibits the positive effect of humanin on insulin sensitivity (PubMed:19623253). Promotes testicular germ cell apoptosis (PubMed:19952275). Acts via LRP-1/alpha2M receptor, also known as TGF-beta type V receptor, to mediate cell growth inhibition independent of IGF1 (PubMed:9252371). Mechanistically, induces serine-specific dephosphorylation of IRS1 or IRS2 upon ligation to its receptor, leading to the inhibitory cascade (PubMed:15371331). In the nucleus, interacts with transcription factors such as retinoid X receptor-alpha/RXRA to regulate transcriptional signaling and apoptosis (PubMed:10874028).

How the assay works

Insulin like growth factor binding protein 3 (IGFBP3) belongs to the family of insulin like growth factor (IGF) binding proteins. IGFBP3 is the main IGF binding protein in plasma, and forms a complex with IGF-I or IGF-II and an acid-labile subunit. IGFBP3 is expressed in multiple tissues, where together with IGFs, they play important roles in glucose homeostasis, cell survival, cell growth and differentiation. The concentrations of IGFBP3 are altered in disease states like type I diabetes, where urinary levels are increased during microalbuminuria, and in prostate cancer, where IGFBP3 protein levels decrease.

Interpolated concentrations of native IGFBP3 in human serum and plasma samples.
The concentrations of IGFBP3 were measured in duplicates, interpolated from the IGFBP3 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:300, plasma (citrate) 1:300, plasma (heparin) 1:333, and plasma (EDTA) 1:500. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP3 concentration was determined to be 2,161 ng/mL in serum, 2,175 ng/mL in plasma (citrate), 822 ng/mL in plasma (heparin), and 1,944 ng/mL in plasma (EDTA).
Interpolated concentrations of native IGFBP3 in SH-SY5Y supernatant samples.
The concentrations of IGFBP3 were measured in duplicates, interpolated from the IGFBP3 standard curves and corrected for sample dilution. Undiluted samples are as follows: SH-SY5Y supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP3 concentration was determined to be 4.28 ng/ml in SH-SY5Y supernatant.
Serum from ten individual healthy human male donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP3 concentration was determined to be 1442 ng/mL with a range of 104 – 3,095 ng/mL.
