Interpolated concentrations of human IgG3 in urine, saliva, and milk. Interpolated concentration of native IgG3 was measured in duplicate at different sample concentrations. Cross reactivity of related immunoglobulin proteins. 50 ng/mL of natively purified human IgG1 IgG2, IgG4, IgA, IgD, IgE, and IgM were tested individually for cross-reactivity. Percent reactivity was determined by interpolating the protein concentration from the human IgG3 standard curve and expressing the interpolated concentration as a percentage of the protein concentration tested.

Interpolated concentrations of IgG3 in normal human serum donors. Serum of ten individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IgG3 concentration was determined to be 340 µg/mL with a range of 163 – 850 µg/mL.

How the assay works

Human IgG3 ELISA Kit, Chemiluminescent (ab325734) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Constant region of immunoglobulin heavy chains. Immunoglobulins, also known as antibodies, are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound immunoglobulins serve as receptors which, upon binding of a specific antigen, trigger the clonal expansion and differentiation of B lymphocytes into immunoglobulins-secreting plasma cells. Secreted immunoglobulins mediate the effector phase of humoral immunity, which results in the elimination of bound antigens (PubMed : 20176268, PubMed : 22158414). The antigen binding site is formed by the variable domain of one heavy chain, together with that of its associated light chain. Thus, each immunoglobulin has two antigen binding sites with remarkable affinity for a particular antigen. The variable domains are assembled by a process called V-(D)-J rearrangement and can then be subjected to somatic hypermutations which, after exposure to antigen and selection, allow affinity maturation for a particular antigen (PubMed : 17576170, PubMed : 20176268).
