Interpolated concentrations of human IKK gamma/NEMO in Jurkat and HeLa cell culture extract.

The endogenous concentration of IKK gamma/NEMO was measured in duplicate at different sample concentrations. Undiluted samples are as follows : Jurkat 50 μg/mL and HeLa 100 μg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Human IKK gamma/NEMO ELISA kit (ab284617) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human IKK gamma/NEMO protein in human cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human IKK gamma/NEMO with 159 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Regulatory subunit of the IKK core complex which phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor (PubMed : 14695475, PubMed : 20724660, PubMed : 21518757, PubMed : 9751060). Its binding to scaffolding polyubiquitin plays a key role in IKK activation by multiple signaling receptor pathways (PubMed : 16547522, PubMed : 18287044, PubMed : 19033441, PubMed : 19185524, PubMed : 21606507, PubMed : 27777308, PubMed : 33567255). Can recognize and bind both 'Lys-63'-linked and linear polyubiquitin upon cell stimulation, with a much higher affinity for linear polyubiquitin (PubMed : 16547522, PubMed : 18287044, PubMed : 19033441, PubMed : 19185524, PubMed : 21606507, PubMed : 27777308). Could be implicated in NF-kappa-B-mediated protection from cytokine toxicity. Essential for viral activation of IRF3 (PubMed : 19854139).
