Product details

Human IL-1 alpha ELISA Kit ab100560 is a sandwich ELISA to measure Human IL-1 alpha in serum, plasma, cell culture supernatant with a sensitivity of 2 pg/ml.

How the assay works

This assay employs an antibody specific for Human IL-1 alpha coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-1 alpha present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-1 alpha antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1 alpha bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm

1. Add standard or sample to each well used. Incubate at room temperature
2. Wash and add prepared biotinylated antibody to each well. Incubate at room temperature.
3. Wash and add prepared streptavidin solution. Incubate at room temperature.
4. Add TMB Development Solution to each well. Incubate at room temperature.
5. Add Stop Solution to each well. Read at 450nm immediately.

Target data

Cytokine constitutively present intracellularly in nearly all resting non-hematopoietic cells that plays an important role in inflammation and bridges the innate and adaptive immune systems (PubMed : 26439902). After binding to its receptor IL1R1 together with its accessory protein IL1RAP, forms the high affinity interleukin-1 receptor complex (PubMed : 17507369, PubMed : 2950091). Signaling involves the recruitment of adapter molecules such as MYD88, IRAK1 or IRAK4 (PubMed : 17507369). In turn, mediates the activation of NF-kappa-B and the three MAPK pathways p38, p42/p44 and JNK pathways (PubMed : 14687581). Within the cell, acts as an alarmin and cell death results in its liberation in the extracellular space after disruption of the cell membrane to induce inflammation and alert the host to injury or damage (PubMed : 15679580). In addition to its role as a danger signal, which occurs when the cytokine is passively released by cell necrosis, directly senses DNA damage and acts as a signal for genotoxic stress without loss of cell integrity (PubMed : 26439902).
