Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3 (PubMed:16982608). In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators (PubMed:18025162). Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF-alpha (PubMed:11564774, PubMed:1940799, PubMed:7512027). Interferes also with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation (PubMed:8144879). In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling (By similarity).

Abcam's IL-10 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-10 in serum, plasma and cell culture supernatants.
This assay employs an antibody specific for Human IL-10 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-10 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

IL-10 measured in culture supernatants from control (-) or activated cell lines (dilution range 1/1-1/00), stimulated for 24 hours with 50 ng x mL-1 of PMA (P)(Phorbol 12-myristate 13-acetate (PMA), PKC activator ab120297) and with the addition of 1 ug x mL-1 of LPS (Sigma) (P/L) for the last 6 hours or in PBMC supernatants (dilution range 1/10-1/1000), from control or treated cells ((PHA) 48 hours with 2% PHA-M; LifeTechnologies) (duplicates +/- SD).
