Interpolated concentrations of native IL-10 in human PBMC cell culture supernatant samples.

The concentrations of IL-10 were measured in duplicates, interpolated from the IL-10 standard curves and corrected for sample dilution. Undiluted samples are as follows : stimulated PBMC cell culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-10 concentration was determined to be 462 pg/ml in stimulated PBMC cell culture supernatant. PBMC cell culture supernatant samples were stimulated for 46 hours with 1.5% PHA. Unstimulated PBMC cell culture supernatant samples were also measured; all values were below the detectable range of the assay.

IL-10 in vitro CatchPoint SimpleStep ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IL-10 protein in human serum, plasma, and cell culture supernatant samples

The CatchPoint SimpleStep ELISA employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. CatchPoint HRP Development Solution containing the Stoplight Red Substrate is added. During incubation, the substrate is catalyzed by HRP generating a fluorescent product. Signal is generated proportionally to the amount of bound analyte and the intensity is measured in a fluorescence plater reader at 530/570/590 nm Excitation/Cutoff/Emission.

Interleukin 10 (IL-10) is a cytokine involved in immunoregulation through the promotion of Th2 response and inhibition of Th1 responses. It is produced by cells of hematopoietic origin, keratinocytes, placental cytotrophoblasts, and hepatic stellate cells.

Target data

Major immune regulatory cytokine that acts on many cells of the immune system where it has profound anti-inflammatory functions, limiting excessive tissue disruption caused by inflammation. Mechanistically, IL10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB leading to JAK1 and STAT2-mediated phosphorylation of STAT3 (PubMed : 16982608). In turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators (PubMed : 18025162). Targets antigen-presenting cells (APCs) such as macrophages and monocytes and inhibits their release of pro-inflammatory cytokines including granulocyte-macrophage colony-stimulating factor /GM-CSF, granulocyte colony-stimulating factor/G-CSF, IL-1 alpha, IL-1 beta, IL-6, IL-8 and TNF (PubMed : 11564774, PubMed : 1940799, PubMed : 7512027). Also interferes with antigen presentation by reducing the expression of MHC-class II and co-stimulatory molecules, thereby inhibiting their ability to induce T cell activation (PubMed : 8144879). In addition, controls the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling (By similarity).
