Interpolated concentration of native IL-11 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : 15% treated MCR-5 cell culture supernatant (treated with 10 ng/mL PMA + 1 ng/mL IL-1 alpha for 48 hrs) and 60% untreated MCR-5 cell culture supernatant. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS. No signal was observed in neat cell culture media (DMEM containing 10% fetal bovine serum).

Human IL-11 ELISA Kit, Chemiluminescent (ab326005) is a rapid single-wash sandwich 55-min ELISA to measure Human IL-11 in Serum, Citrate Plasma, EDTA Plasma, Cell Culture Supernatant with a sensitivity of 4.5 pg/mL.

How the assay works

Human IL-11 ELISA Kit, Chemiluminescent (ab326005) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Cytokine that stimulates the proliferation of hematopoietic stem cells and megakaryocyte progenitor cells and induces megakaryocyte maturation resulting in increased platelet production (PubMed : 2145578). Also promotes the proliferation of hepatocytes in response to liver damage. Binding to its receptor formed by IL6ST and IL11RA activates a signaling cascade that promotes cell proliferation (PubMed : 12919066). Signaling leads to the activation of intracellular protein kinases and the phosphorylation of STAT3. The interaction with the membrane-bound IL11RA and IL6ST stimulates 'classic signaling', whereas the binding of IL11 and soluble IL11RA to IL6ST stimulates 'trans-signaling' (PubMed : 30279168).
