Interpolated concentrations of native IL-15 in human heart homogenate tissue extract sample based on a 250 μg/mL extract load.

The concentrations of IL-15 were measured in duplicate and interpolated from the IL-15 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-15 concentration was determined to be 22.5 pg/mL in human heart homogenate tissue extract.

How the assay works

Human IL-15 SimpleStep ELISA^{^®}employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^{^®} plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature

IL-15 (114 amino acids) is a cytokine that regulates T and natural killer cell activation and proliferation. It has a predicted molecular mass of approximately 12.5 kDa. IL-15 was initially isolated from the simian kidney epithelial cell line CV1/EBNA. It has also been isolated from mouse and human cell sources. The cytokines IL-15 and IL-2 share many biological properties and stimulatory activities (T, B, and NK cells). There is no sequence homology between IL-15 and IL-2, though computer modeling indicates both possess a four alpha helical bundle structure. IL-15 competes for binding sites with IL-2, as both IL-2 and IL-15 stimulate the growth of cells through the IL-2 receptor. IL-15 mRNA is expressed in many cell types and tissues including adherent peripheral blood mononuclear cells, fibroblasts, and epithelial cells, monocytes, placenta, and skeletal muscle.

Cytokine that plays a major role in the development of inflammatory and protective immune responses to microbial invaders and parasites by modulating immune cells of both the innate and adaptive immune systems (PubMed : 15123770). Stimulates the proliferation of natural killer cells, T-cells and B-cells and promotes the secretion of several cytokines (PubMed : 8178155, PubMed : 9326248). In monocytes, induces the production of IL8 and monocyte chemotactic protein 1/CCL2, two chemokines that attract neutrophils and monocytes respectively to sites of infection (PubMed : 9326248). Unlike most cytokines, which are secreted in soluble form, IL15 is expressed in association with its high affinity IL15RA on the surface of IL15-producing cells and delivers signals to target cells that express IL2RB and IL2RG receptor subunits (PubMed : 10233906, PubMed : 23104097, PubMed : 8026467). Binding to its receptor triggers the phosphorylation of JAK1 and JAK3 and the recruitment and subsequent phosphorylation of signal transducer and activator of transcription-3/STAT3 and STAT5 (PubMed : 7568001). In mast cells, induces the rapid tyrosine phosphorylation of STAT6 and thereby controls mast cell survival and release of cytokines such as IL4 (By similarity).
