Serum of ten individual healthy human male donors and two individual human female donors diagnosed with rheumatoid arthritis was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean IL-15RA concentration was determined to be 205.6 pg/mL for normal male donors and 346.3 pg/mL for rheumatoid arthritis female donors.

The concentrations of IL-15RA were measured in duplicates interpolated from the IL-15RA standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50% and plasma (citrate) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean IL-15RA concentration was determined to be 170.6 pg/mL in serum and 157.8 pg/mL in plasma (citrate).

The concentrations of IL-15RA were measured in duplicates interpolated from the IL-15RA standard curves and corrected for sample dilution. Undiluted samples are as follows : unstimulated PBMCs 50% and stimulated PBMCs 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean IL-15RA concentration was determined to be undetectable in unstimulated PBMCs and 90.8 pg/mL in stimulated PBMCs.

How the assay works

Human IL-15RA ELISA Kit, Chemiluminescent (ab326308) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

High-affinity receptor for interleukin-15 (PubMed : 8530383). Can signal both in cis and trans where IL15R from one subset of cells presents IL15 to neighboring IL2RG-expressing cells (By similarity). In neutrophils, binds and activates kinase SYK in response to IL15 stimulation (PubMed : 15123770). In neutrophils, required for IL15-induced phagocytosis in a SYK-dependent manner (PubMed : 15123770).
