Product details

Abcam's IL-17 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-17 in plasma and cell culture supernatants.

This assay employs an antibody specific for Human IL-17 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-17 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-17 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-17 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

We have not been able to detect endogenous Human IL-17 in normal serum with ab100556, only in serum spiked with Human IL-17.

Optimization may be required with urine samples

Effector cytokine of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity (PubMed : 24120361). Signals via IL17RA-IL17RC heterodimeric receptor complex, triggering homotypic interaction of IL17RA and IL17RC chains with TRAF3IP2 adapter. This leads to downstream TRAF6-mediated activation of NF-kappa-B and MAPkinase pathways ultimately resulting in transcriptional activation of cytokines, chemokines, antimicrobial peptides and matrix metalloproteinases, with potential strong immune inflammation (PubMed : 17911633, PubMed : 18684971, PubMed : 19825828, PubMed : 21350122, PubMed : 24120361, PubMed : 8676080). Plays an important role in connecting T cell-mediated adaptive immunity and acute inflammatory response to destroy extracellular bacteria and fungi. As a signature effector cytokine of T-helper 17 cells (Th17), primarily induces neutrophil activation and recruitment at infection and inflammatory sites (By similarity). In airway epithelium, mediates neutrophil chemotaxis via induction of CXCL1 and CXCL5 chemokines (By similarity). In secondary lymphoid organs, contributes to germinal center formation by regulating the chemotactic response of B cells to CXCL12 and CXCL13, enhancing retention of B cells within the germinal centers, B cell somatic hypermutation rate and selection toward plasma cells (By similarity).
