Human IL-2 ELISA Kit is a single-wash, 90-minute SimpleStep ELISA® used to quantify human interleukin-2 (IL-2) with a sensitivity of 32.1 pg/mL. The assay employs a simple Mix-Wash-Read protocol with just one incubation and wash step.

Linearity of dilution is determined based on interpolated values from the standard curve. Linearity of dilution defines a sample concentration interval in which interpolated target concentrations are directly proportional to sample dilution.

Recombinant IL-2 was spiked into the following biological samples and diluted in a 2-fold dilution series in Sample Diluent 50BP.

50% pooled serum samples from eight healthy donors was measured in duplicate. All values were below the detectable range of the assay.

Interpolated concentrations of native IL-2 in human stimulated PBMCs.

The concentrations of IL-2 were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : PBMCs stimulated with 1.5% PHAM for 48 hours (25%). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 5,330 pg/mL in stimulated PBMCs.

Human IL-2 ELISA Kit ab270883 is a rapid single-wash 90-min ELISA kit to measure Human IL-2 in cell culture media, cell culture supernatant, serum and plasma. This SimpleStep ELISA® sensitivity is 32.1 pg/mL.

How the assay works
Human IL-2 SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit recognizes both native and recombinant human IL-2 protein in stimulated serum, plasmas, and cell culture supernatant. Urine, milk, saliva, and cell and tissue extract samples have not been tested with this kit. Recombinant human IL-2R alpha was prepared at 5,000 pg/mL and assayed for cross reactivity. No cross-reactivity was observed. Recombinant human IL-2R alpha was prepared at 5,000 pg/mL and tested for interference. No interference with was observed.

Target data

Cytokine produced by activated CD4-positive helper T-cells and to a lesser extend activated CD8-positive T-cells and natural killer (NK) cells that plays pivotal roles in the immune response and tolerance (PubMed : 6438535). Binds to a receptor complex composed of either the high-affinity trimeric IL-2R (IL2RA/CD25, IL2RB/CD122 and IL2RG/CD132) or the low-affinity dimeric IL-2R (IL2RB and IL2RG) (PubMed : 16293754, PubMed : 16477002). Interaction with the receptor leads to oligomerization and conformation changes in the IL-2R subunits resulting in downstream signaling starting with phosphorylation of JAK1 and JAK3 (PubMed : 7973659). In turn, JAK1 and JAK3 phosphorylate the receptor to form a docking site leading to the phosphorylation of several substrates including STAT5 (PubMed : 8580378). This process leads to activation of several pathways including STAT, phosphoinositide-3-kinase/PI3K and mitogen-activated protein kinase/MAPK pathways (PubMed : 25142963). Functions as a T-cell growth factor and can increase NK-cell cytolytic activity as well (PubMed : 6608729). Promotes strong proliferation of activated B-cells and subsequently immunoglobulin production (PubMed : 6438535). Plays a pivotal role in regulating the adaptive immune system by controlling the survival and proliferation of regulatory T-cells, which are required for the maintenance of immune tolerance. Moreover, participates in the differentiation and homeostasis of effector T-cell subsets, including Th1, Th2, Th17 as well as memory CD8-positive T-cells.
